Vaccine-induced memories

Epidemiological studies initially revealed that certain live attenuated vaccines, such as the tuberculosis vaccine, BCG, the oral polio vaccine, the measles vaccine, or the smallpox vaccine, provide protection beyond their targeted pathogens, decreasing overall mortality and morbidity. Several mechanisms have been proposed to explain these heterologous non-specific effects (NSE) of vaccines, including the induction of cross-reactive B and T cells, the bystander activation of specific T cells, and the establishment of an innate immune memory conferring a non-specific trained immunity.
We are investigating the mechanisms leading to trained immunity in non-human primates immunized with the smallpox and Mpox vaccine, MVA, and with BCG, in collaboration with Roland Brosch. We study blood myeloid cells and myelopoeisis. Deciphering the mechanisms at play should contribute to harness innate memory to improve responses to vaccines.
