NLRP3 comes in different flavours
NLRP3 is a pattern recognition receptor well-known for its role in the inflammasome assembly upon danger or pathogen sensing in myeloid cells. This complex plays crucial role in the maturation of inflammatory cytokines and in the induction of a lytic cell death named pyroptosis. Excessive inflammasome activation results in inflammatory conditions, and autosomal dominant mutations cause NLRP3-associated autoinflammatory disorders. But NLRP3 is not only involved in inflammasome assembly and pathogen recognition. We discovered it is a crucial actor of the maintenance of genome stability. In this seminar, I will discuss new mechanisms keeping the NLRP3 inflammasome on check and novel nuclear functions of NLRP3, independent of the inflammasome, in the repair of DNA double strand break.
