Notre équipe combine des techniques de pointe en imagerie optique et électronique, en science des matériaux, en mécanique cellulaire et en imagerie intravitale pour élucider la manière dont les phagocytes, en particulier les macrophages et les ostéoclastes, interagissent avec la matrice extracellulaire. L’objectif est de déchiffrer les mécanismes de migration des macrophages en 3D et d’étudier comment le VIH-1 manipule la propagation des phagocytes d’une cellule à l’autre.
À long terme, nos projets permettront d’identifier de nouvelles cibles pour contrôler les interactions des macrophages avec leur environnement, ce qui sera utile dans la lutte contre le cancer et l’infection par le VIH.
Nous étudions comment les phagocytes interagissent avec leur microenvironnement, dans des contextes physiologiques et pathologiques, y compris le cancer et les maladies infectieuses.
Les macrophages sont des cellules immunitaires innées qui migrent et sont présentes dans tous les tissus pour maintenir la surveillance immunitaire. Ils sont la cible de pathogènes et ingèrent des particules telles que des bactéries ou des cellules mortes, par un processus appelé phagocytose. Notre équipe se concentre sur les mécanismes utilisés par les macrophages pour interagir avec leur environnement, afin de mettre en évidence des molécules qui pourraient être utilement ciblées pour limiter leur action délétère.
En utilisant la microscopie intravitale, notre équipe a révélé que les macrophages utilisent le mode de migration mésenchymateuse pour infiltrer les tumeurs denses et que les inhibiteurs des métalloprotéases matricielles diminuent à la fois le nombre de macrophages associés à la tumeur et la croissance tumorale (Gui et al. 2018 Cancer Immunol Res). Les podosomes sont des structures d’adhérence cellulaire impliquées dans la dégradation de la matrice extracellulaire et la migration mésenchymateuse des macrophages. Ils sont composés d’un noyau submicronique de filaments d’actine entouré d’un anneau d’adhérence constitué entre autres d’intégrines.
Grâce à une méthode que nous avons appelée microscopie à force de protrusion, nous avons démontré que les podosomes génèrent des forces de protrusion proportionnelles à la rigidité de la matrice extracellulaire (Labernadie et al. 2014 Nat Commun ; Proag et al. 2015 ACS Nano ; Labernadie et al. 2022 Nat Commun), et impliquent un équilibre des forces entre la protrusion du cœur d’actine et une traction au niveau de l’anneau d’adhérence (Bouissou et al. 2017 ACS Nano). Dans les ostéoclastes dégradant les os, les podosomes s’assemblent en une super structure appelée zone de scellement. Nous avons montré à l’aide de techniques de microscopie à super-résolution qu’il existe une coordination locale des cœurs de podosomes au sein d’îlots à l’échelle micrométrique (Portes et al. 2022 Elife). Nous avons également développé un dispositif combinant des microcanaux et des piliers et avons montré que les forces sont redirigées de l’intérieur vers l’extérieur lorsque le confinement cellulaire augmente (Desvignes et al. 2018 Nano Lett). Plus récemment, nous avons démontré, en étroite collaboration avec Marion Jasnin et Serge Dmitrieff, que les forces, à l’échelle nanométrique, exercées par les podosomes peuvent être expliquées par l’énergie élastique stockée dans les réseaux d’actine des podosomes (Jasnin et al. 2022 Nat Commun).
Nous avons montré que la protéine Nef du VIH-1 module la migration des macrophages à la fois in vitro et in vivo, et favorise la dissémination du virus en augmentant la migration mésenchymateuse et en modulant la structure et la fonction des podosomes (Vérollet et al. 2015 Blood). De plus, nous avons observé que les ostéoclastes sont infectés de manière productive par le VIH-1. Le virus altère fortement l’organisation des podosomes dans les ostéoclastes, conduisant à une activité de résorption osseuse accrue (Raynaud-Messina et al. 2018 PNAS). Ces observations expliquent probablement l’accumulation de macrophages dans plusieurs tissus des patients infectés par le VIH-1 et pourquoi ils souffrent d’ostéolyse. Les macrophages sont également les principales cellules hôtes de Mycobacterium tuberculosis (Mtb). Dans le contexte de la tuberculose, nous avons montré que la migration mésenchymateuse des macrophages est renforcée et associée à une accumulation de macrophages sensibles à Mtb dans les poumons (Lastrucci et al. 2015 Cell Res). Nous avons également montré que, dans le microenvironnement de la tuberculose, la formation de nanotubes (TNT) par les macrophages est augmentée. Lorsque ces macrophages sont ensuite infectés par le VIH-1, le virus se propage entre les cellules en utilisant les TNT et la lectine Siglec-1/CD169. Ces mécanismes pourraient expliquer comment la tuberculose renforce la pathogenèse du VIH-1 chez les patients co-infectés (Souriant et al. 2019 Cell Rep ; Dupont et al. 2020 Elife ; Dupont et al. 2022 J Leuk Biol). Plus récemment, nous avons révélé un mécanisme nouveau et efficace d’infection des macrophages tissulaires par le VIH-1 via la fusion avec des lymphocytes T CD4 infectés (Mascarau et al. 2023 J Cell Biol).

In our article published in the EMBO Journal, we demonstrate that macrophage migration can occur without the ERM proteins, which were previously considered essential for any cell migration. Their results show that specific mechanisms are at work in these key cells of our immune system.
